چکیده

Reduced glutathione (GSH) plays a crucial role in ensuring that other thiol groups remain reduced within the cell and is particularly important in the biosynthesis of DNA [for ;I review, see Holmgren ( 1985)l. Dihydrolipoamide dehydrogenase is an essential component of the 2-ox0 acid dehydrogenase multi-enzyme complexes (Reed, 1974; Perham, 1983) in which it acts to oxidize the dihydrolipoic acid residues of the lipoate acyltransferase components in an NAD + -dependent reaction: Lip(SH), + NAD' = Lips2 + NADH + H'

Protein engineering of domains in flavoprotein disulphide oxidoreductases: contributions to folding and assembly.

Introduction The flavoprotein disulphide oxidoreductases constitute a growing family of homologous dimeric enzymes, with an important and diverse range of functions in vivo. Among its members are dihydrolipoyl dehydrogenase, glutathione reductase, mercuric reductase, thioredoxin reductase, trypanothione reductase and a related enzyme, NADPH peroxidase (for recent reviews of their structures and...

Glutathione-dependent activities of Trypanosoma cruzi p52 makes it a new member of the thiol:disulphide oxidoreductase family.

Trypanothione: glutathione disulphide thioltransferase of Try-panosoma cruzi (p52) is a key enzyme in the regulation of the intracellular thiol-disulphide redox balance by reducing glutathione disulphide. Here we show that p52, like other disulphide oxidoreductases possessing the CXXC active site motif, catalyses the reduction of low-molecular-mass disulphides (hydroxyethyl-disulphide) as well ...

A glutathione reductase from Escherichia coli.

Earlier studies had demonstrated that a flavoprotein capable of mediating a triphosphopyridine nucleotide (TPN)-linked reduction of Furacin (5-nitro-2-furaldehyde semicarbazone) was present in cell-free extracts of Furacin-susceptible strains of Escherichia coli, but was absent in extracts of resistant strains (I). Dr. Philip E. Hartman, who was then associated with our department, suggested to...
